摘要
目的研究检测血清C1q抗体方法。方法建立ELISA检测血清C1q抗体水平的程序,应用棋盘滴定的方法确定ELISA检测人血清C1q抗体系统中包被抗原以及酶标抗体的最适工作浓度。检测92份正常健康体检合格者的血清C1q抗体水平,确定参考值范围。结果C1q抗原的包被浓度5μg/ml,酶标抗体的工作浓度1∶50000。批内和批间重复性试验的变异系数均小于<15%。血清C1q抗体浓度参考值范围0-30.55 U/ml。结论小批血清的实验检测表明此方法的特异性较好,操作简单。适于血清C1q抗体水平的检测。
Objective To study the method of detecting the serum level of anti-Clq antibody.Methods Develop the procedure of detecting the serum level of anti-Clq antibody with EHSA. And among the anti-Clq antibody ELISA detection system,the concentration of the coated antigen and the enzyme labeled antibody is determined by the checkerboard titration technique. Finally sera from 92 healthy people were tested by the anti-Clq antibody ELISA detection system to decide the reference value.Results The concentration of the coated antigen is 5μg/ml, and the concentration of the enzyme labeled antibody is 1： 50000. And coefficient of variability percent （CV %） of intro-batch and inter-batch reproducibility test was less than 15 %. According the result of the 92 people, the reference value is determined. [0-30.55 U/ml] Conclusion The specificity of the anti-Clq antibedy ELISA detection system was approved nice by detecting small amounts of sera.And the procedure is not complicated. The anti-Clq antibody ELISA detection system is good for the serum level of anti-Clq antibody.
