摘要
用RAPD技术分析了广东养殖大口黑鲈Micropterus salmoides3个群体基因组DNA的多态性。结果表明:用12条随机引物对3个群体共88尾大口黑鲈的基因组DNA进行扩增,共获得了106个位点,平均每条随机引物扩增出8.8个位点;广州群体、顺德大良群体和顺德南水群体的多态位点比例分别为32.08%、33.02%和40.57%;Nei遗传多样性指数分别为0.1151、0.1176和0.1627;Shannon s遗传多样性指数分别为0.1708、0.1742和0.2378。这表明目前广东地区养殖大口黑鲈种质资源还具有一定的遗传多样性,但养殖群体的遗传多样性在降低,且具有不同程度的种质退化。
Random amplified polymorphic DNA (RAPD) method was used to detect the genetic diversity of cultured largemouth bass Micropterus salmoides obtained from Guangdong province. Using 12 random primers, 106 RAPD sites were detected, with the average loci of 8.8 in each primer. The Guangzhou, Shunde and Nanshui pop- ulations had mean polymorphic loci percentage of 32.08% ,33.02% and 40.57% ; Nei's gene diversity of0.1151, 0.1176 and 0. 1627 and the Shannon's diversity of 0. 1708,0. 1742 and 0.2378, respectively. These indexes indicate that the cultured largemouth bass from Guangdong province has not decrease in genetic diversity at present, but in some areas has decrease.
