苹果新品种自交不亲和基因型的分子生物学鉴定 被引量:3

摘要 根据保守氨基酸序列FTQQYQ和anti-1/MIWPNV设计苹果自交不亲和基因引物P1:5′-TT-TACGCAGCAATATCAG-3′;P2:5′-ACGTTCGGCCAAATA/CATT-3′,利用PCR-RFLP分析方法,以河北省新引进的苹果品种美国8号、昂林、松本锦、华冠、南方脆幼叶为试验材料,鉴定其自交不亲和基因型分别为S9S27、S1S9、S9S9、S2S9、S2S9。 Five new apple cultivars, USA No. 8, Korin, Matsumoto Nishiki, Huaguan and Nanfangcui, which were introduced into Hebei Province recently, were used as materials in the present study. The primers ( Pi : 5'-TTTACGCAGCAATATCAG-3' ; P2 : 5'-ACGTrCGGCCAAATA/CATT-3') were newly designed according to the conserved sequence of amino acid of S-RNase. The self-incompatibility alleles of the cultivars were isolated and identified using PCR - RFLP analysis. Based on the results, the self-incompatibility genotypes of the cuhivars were identified as S9S27, S1S9, S9S9, S2S9 and S2S9, respectively.
