已酮可可碱对内毒素血症大鼠肝损伤的保护作用 被引量:3

Protective effects of pentoxifylline on hepatic tissue in rats with endotoxemia

摘要 目的探讨已酮可可碱(PTX)对内毒素血症大鼠肝损伤的保护作用。方法Wistar大鼠18只随机分成三组:假手术组、内毒素组和PTX治疗组,每组6只。测定各组动物血清TNF-α、IL-1β水平,肝组织ICAM-1蛋白表达。结果内毒素组血清TNF-α和IL-1β水平与假手术组比较明显增强(P<0.01),肝组织ICAM-1蛋白表达与假手术组比较也明显增强(P<0.01)。PTX治疗后,血清TNF-α、IL-1β水平和肝组织ICAM-1蛋白表达与内毒素组比较明显减弱(P<0.01)。结论TNF-α,IL-1β和肝组织ICAM-1在内毒素急性肝损伤发病机制中起重要作用,PTX可以作用于TNF-α、IL-1β和肝ICAM-1,下调其表达,减轻内毒素对肝组织造成的损伤,保护肝组织。 Objective To study the protective effects of pentoxifylline on hepatic tissue in rats with endotoxemia. Methods 18 Wistar rats were randomly divided into 3 groups:sham operated control group( S group), LPS group and PTX group. The serum tumor necrosis factor-α ( TNF -α) and interleulein - 1 β ( IL - 1β) levels in each groups were measured. The immunohistochemistry was used to assess the expression of the ICAM - 1 in the hepatic tissues. Results The levels of TNF - α and IL - 1β in serum in LPS group were higher than those in S group ( P 〈 0.01 ). The levels of ICAM - 1 in the hepatic tissues in L groups were higher than those in sham operated control group ( P 〈 0.01 ). After PTX treatment, the levels of TNF - α and IL - 1 β in serum in PTX group were lower than those in LPS group ( P 〈 0.01 ) and the levels of ICAM - 1 in the hepatic tissues in PTX group were also lower than those in LPS group ( P 〈 0. 01 ). Conclusion TNF - α, IL - 1 β and ICAM - 1 take part in the progress of hepatic injury in rats with endotoxemia. PTX can eliminate the hepatic injury and protect the hepatic tissues by downregulating the levels of TNF - α, IL - 1 β and ICAM - 1.
