摘要
基于色氨酸和半胱氨酸对Ru(pheo)3^2+-Ce(Ⅳ)体系化学发光的增强作用及其发光动力学性质的显著差别,提出了一种化学发光法同时测定这两种氨基酸的新方法。在优化的实验条件下,该方法测定色氨酸和半胱氨酸的线性范围分别为0.05~2.0μg/mL和0.05~3.0μg/mL,检出限分别为0.03μg/mL和0.02μg/mL。将其应用于合成样品中两种氨基酸的同时测定。结合文献提出了可能的化学发光反应机理。
It was observed that both tryptophan and cysteine could enhance the chemiluminescence (CL) intensity of tris-(1,10-phenanthroline) ruthenium ( Ⅱ ) (Ru(phen)3^2+ ) - Ce(Ⅳ) system and their CL kinetics curves were remarkably different. Based on above phenomena, a novel CL method was proposed for the simultaneous determination of tryptophan and cysteine. Under the optimum experimental conditions, the linear range and detection limit for tryptophan were 0.05 - 2.0μg/mL and 0.03μg/mL, respectively; and 0.05 - 3.0μg/mL and 0.02μg/mL for cysteine, respectively. The proposed method has been used to analyze the synthetic samples. The possible mechanism of CL reaction was studied.
