摘要 目的比较粘着斑激酶(FAK)和整合素(integrin)α5β1在大肠癌及癌旁正常组织中的表达与肿瘤细胞分化、浸润、转移等生物学行为的关系。方法用免疫组织化学SP法检测60例大肠癌及癌旁组织中FAK和整合素α5β1蛋白的表达水平。结果大肠癌组织FAK和整合素α5β1的表达均明显高于癌旁组织(P<0.01);低分化癌组织FAK和整合素α5β1的表达均高于高、中分化癌组织(P<0.05);FAK表达与癌组织浸润深度及淋巴结转移有关(P<0.01);而整合素α5β1表达与癌组织浸润深度及淋巴结转移无关(P>0.05)。结论FAK较整合素α5β1可以更有效地衡量大肠癌的发生与发展,FAK的表达水平可作为大肠癌发生、发展过程中1种较有价值的指标。 Objective To compare the expression of focal adhesion kinase (FAK) and integrin ct5 61 in colorectal carcinoma (CRC) and non-cancerous tissue and study their relations with clinicopathological data of the tumor. Methods The expression levels of FAK and integrin α5β1were detected in 60 CRCs by using an immunohistochemical technique. Results The expression of FAK and integrin α5β1 in CRC tissues was significantly higher than that in non-cancerous tissues ( P 〈 0.01 ). The FAK and integrinα5β1 lexpression in poor differentiated tissues was higher than that in well-moderate differentiated tissues (P 〈 0.05). The FAK expression was correlated with the infiltration depth and metastasis of CRC (P 〈 O. 01 ), whereas the integrinα5β1 expression was not correlated with the infiltration depth and metastasis of CRC (P 〉 0.05 ). Conclusion FAK is a more significant indicator than integrin α5β1 in evaluating carcinogenesis and progression of the CRC. The level of FAK expression might be used as a valuable marker for the carcinogenesis and progression of the CRC.
