摘要
制备大鼠肝抑素(HC)纯品,研究它对再生肝细胞和肝癌细胞体外增殖的影响。用DEAE-Cellulose阴离子交换层析法提取HC纯品,用液法检测HC样品的生物活性。结果:提取的HC纯品的SDS电泳纯度达89.5%, Mr 13.5 × 103;该 HC纯品对大鼠再生肝细胞和人肝癌细胞体外增殖有明显抑制作用,抑制率分别为63.1%(0.2 mg/L)和77.7%(0.3 mg/L),但对人胃癌细胞增殖无抑制效应。结论:结果提示提取的HC是抑制再生肝细胞和肝癌细胞增殖的特异性因子。
PURPOSE To purify rat hepatic chalone, and study its effects in vitro on the proliferation of regenerating hepatocytes and hepatoma cells. METHODS The DEAE-cellulose anion exchange chromatography was used to purify hepatic chalone sample. The fluid scintillation was used to assay the biological activity of hepatic chalone sample. RESULTS The purified hepatic chalone is of 89.5 % purity and 13.5 x 103 molecular weight. This purified hepatic chalone showed remarkable inhibitory effect on proliferation of regenerating hepatocytes and human hepatoma cells in vitro with the inhibitory rate of 63.1 % (0.2 mg / L) and 77.7 % (0.3 mg / L) respectively, but did not show any inhibitory effect on the proliferation of human gastric carcinoma cells. CONCLUSIONS The purified hepatic chalone is a specific factor which can inhibit the proliferation of regenerating hepatocytes and hepatoma cells.
