摘要
本研究目的是建立并鉴定树突状细胞肉瘤体外传代细胞系。取荷LI瘤615小鼠脾脏进行原代细胞培养,经传代培养后新建立一株体外传代细胞系,经鉴定为树突状细胞肉瘤细胞系,命名为DCS。该细胞体外呈分支状。酶组织化学检查发现碱性磷酸酶、酸性磷酸酶、非特异性脂酶、过氧化酶及胞膜ATPase均阴性。用DCS细胞免疫家兔制备特异抗血清,阳性产物仅见于脾动脉周围的淋巴鞘和淋巴滤泡周边区,呈树枝状,生发中心阴性,皮肤Langerhans细胞呈阳性。初步认为该细胞来源于小鼠脾交错树突状细胞。
DCS cells has been established from splenic tumor of a mouse. The cells is adherent, dendritic, and its cytoplasm has some global mitochondria but few phagolysosomes. DCS does not express differentiation antigens of monocytes, lymphocytes. We used the polyclonal antibodies produced from rabbits against DCS cells to find the original cells in mouse spleen. The positive cells was in the T areas, i.e. the periarteriolar lymphoid sheaths(PAL), and in the skin the positive cells was Langerhans cells. Among adherent splenic cells there were some cells, which did not phagocytose particles and also did not pinocytose HRP, but could be stained by the serum against DCS with the routine ABC method. All cells of DCS was peroxidase negative, acid phosphatase negative,alkaline phosphatase weak or negative,non specific esterases negative and membrane ATPase negative. It is considered that the DCS cells originated from interdigital dendritic cells of mouse spleen.
