摘要 用间接免疫荧光和免疫印迹方法研究CENP-B与HeLa细胞核骨架的关系,并用TritonX-100、Tween-40、脱氧胆酸钠、核酸酶及(NH4)2SO4选择性抽提HeLa细胞,保留细胞非组蛋白支架。ACA间接免疫荧光染色表明在细胞核区域有明亮的荧光斑点。Westernblot分析抽提后的细胞核骨架,发现CENP-B蛋白与细胞核骨架紧密结合。 Centromere kinetochore complex located at the primary constriction region of chromosomes is an important organelle which mediates the attachment of the chromosomes to the spindle microtubules at mitosis and meiosis. CENP B is a highly conserved protein located at the centromere/kinetochore complex. In this paper, the relationship between CENP B and the nuclear skeleton of HeLa cells was investigated. ACA stained HeLa cells after selective extraction with Triton X 100, Tween 40, Sodium Deoxycholate, Nuclease and (NH 4) 2SO 4 and 2M NaCl to keep the intact nuclear skeleton showed bright immunofluorescence spots in nuclear region. Western blot analysis revealed that CENP B protein was closely associated with the nuclear skeleton. It impies that CENP B may be the component of nuclear skeleton and infers that CENP B might directly take part in the formation of chromosomes.
