红色毛癣菌随机扩增DNA多态性分型研究 被引量:9

摘要 目的研究红色毛癣菌DNA分型并探讨DNA分型技术在临床菌种鉴定以及流行病学中的应用。方法氯化苄法提取DNA,并用随机扩增DNA多态性(RAPD)方法对临床分离的30株红色毛癣菌进行DNA分型。结果红色毛癣菌具有独特而稳定的RAPD带型,30株临床分离的红色毛癣菌可进一步分为9型,菌株间相似程度较高。在多数菌株DNA扩增中,红色毛癣菌与有性型Arthrodermabenhamiae和A.vanbreuseghemi均有较大的差异。同一患者不同部位取材的菌株间以及取自不同患者的菌株间DNA型均可相同。结论RAPD方法揭示了红色毛癣菌DNA型的独特性,为红色毛癣菌提供了鉴定与分型的标志,为进一步探讨感染来源。 Objective To investigate the DNA type of Trichophyton rubrum and to discuss the application of DNA typing method in its clinical identification and epidemiology study. Methods DNA extraction was carried out by using benzyl chloride, and random amplification of polymorphic DNA (RAPD) assay was employed for DNA typing of 30 clinical isolates of Trichophyton rubrum. Results T. rubrum showed stable and distinct DNA pattern from other dermatophytes. Thirty clinical isolates of T. rubrum were divided into 9 types, and showed high similarity one another. RAPD also revealed that T. rubrum was different with Arthroderma benhamiae and A. vanbreuseghemii in most amplifications. Identical DNA pattern was found in strains from different sites of infection in the same patient, and even from different patients. Conclusions RAPD shows the distinct DNA type of T. rubrum, and provides markers for classification and identification. It has great significance in investigating the origin, transmission of pathogens and other epidemiology study.
