摘要
利用玻璃纤维丝柱过滤感染弓形虫CFW小鼠的腹腔液,分离纯化速殖子,滤前平均纯度为0.7036±0.164,滤后平均纯度为0.9874±0.014(P>0.01)。研究结果显示用此法分离纯化的速殖子,其毒力与抗原性均不发生改变。将速殖子制备成虫体抗原作斑点酶联免疫吸附试验,检测弓形虫抗体阳性兔血清,呈阳性反应,检测阴性兔血清及阿米巴阳性、球虫阳性和血吸虫阳性兔血清均呈阴性反应,显示了抗原的特异性强。点样于硝酸纤维膜(NC)上的抗原保存于冰箱(-20℃)中一年半后。
In order to prepare high purified Toxoplasma gondii antigen, the peritoneal fluid of infected CFW mice was passed through the glass wool filter to isolate and purify tachyzoites The high purity of 0 987 4±0 014 after treatment versus 0 703 6±0 164 before treatment was obtained (P<0 01) Experimental data showed that the purified tachyzoites remained their proper morphological structure, activity, virulence and antigenicity etc. The results of whole tachyzoite antigen prepared from the purified tachyzoites used in Dot ELISA for detecting the antibodies of toxoplasma gondii confirm that the whole tachyzoite antigen posseses high specificity The antigenicity of the antigen which was frozen in -20℃ without change during 1-1 5 years The preparation of whole thachzote antigen was very simple, and no special reagents and instruments were needed. The preparation of soluble antigen and susceptibility to infection with Toxopleasma Gondii in different strains of mice were also studied in this paper
