摘要 微载体悬浮培养动物细胞条件下,发酵缶的混合与剪切性能优劣对动物细胞的培养效果至关重要。本文研究了NBS动物细胞发酵缶的流体混合时间、搅拌功率和时均速度分布,探讨了微载体及动物细胞所受到的流体作用力的计算方法,为设计大规模动物细胞发酵缶不变提供了部分依据。 Mixing time, power consumption and spatial distribution of time-average velocities were investigated in a fermenter which had been produced by New-Brunswick Sci. Corp.. The working fluid is water. Mixing time was determined by optical fiber probes and power consumption by trunnion dynamometer. The average velocities were measured by DISA 55M hot-film anemometer.It was found that the fermenter could provide efficient mixing and the mixing time could be correlated by equation n θ= 14. 9. On the basis of the experimental data of power consumption and shear rate, the fluid force which might act on mammalian cells and imcrocarriers had been estimated. The force involved are the Reynolds stress owing to the turbulence and the maximum shear stress which resulted from the spatial gradient of average velocity, but the former was suggested to be more important than the latter for damaging mammalian cells and microcarriera.
