气相色谱法测定不同产地白英中亚油酸的含量 被引量:3

摘要 目的:建立气相色谱法测定中草药白英中亚油酸的含量。方法:粉碎后的白英以氯仿为溶剂在索氏提取器中回流提取,提取液经皂化、酯化和正己烷萃取后,采用气相色谱法进行测定。气相色谱条件:HP-5毛细管柱(30 m×0.25 mm×0.25μm),氢火焰离子化检测器(FID),以高纯 N_2为载气。柱温:初始为150℃,保持1 min,以15℃·min^(-1)升温到225℃,再以5℃·min^(-1)升温到260℃,保持3 min。结果:亚油酸甲酯与其他成分分离良好。线性范围为0.02~0.14 mg·mL^(-1)(r=0.9997)。高、中、低3种浓度的平均回收率(n=3)分别为96.5%(RSD=1.9%),96.4%(RSD=1.3%),95.6%(RSD=1.8%)。结论:方法简单、快速、灵敏,重复性好,可用于中草药中亚油酸的含量测定。 Objective:To develop a GC method for the determination of linoleic acid in Solanum Lyratum Thunb. Methods:Solanum Lyratum Thunb was extracted with chloroform by heating reflux. Then, samples were saponified, esterified, extracted with n - hexane and determined by GC. The chromatographic analysis was performed using HP - 5 capillary column (30 m × 0. 25 mm × 0. 25 μm) with FID detector. Highly pure N2 was used as carrier gas. The column temperature was programmed from 150 ℃ (held for 1 min) to 225 ℃ at a rate of 15 ℃· min^-1 ,then to 260 ℃ (held 3 min)at a rate of 5 ℃· min^-1. Results:Methyl linoleate was separated well with other components. There was a good linear relationship within the concentration range of 0.02 - 0. 14 mg · mL ^- 1 ( r = 0. 9997 ). The average recoveries ( n = 3 ) of linoleic acid at high, middle and low concentration were 96.5% ( RSD = 1.9% ), 96. 4% ( RSD = 1.3 % ) and 95.6% ( RSD = 1.8% ) respectively. Conclusions: The method is simple, rapid, accurate and reproducible. It is suitable for the determination of linoleic acid in traditional Chinese herbs.
