地黄饮子对AD模型大鼠的学习记忆及胆碱能损害的影响 被引量:5

Effect of Decoction of Rehmanniae on Improving Learning Memory Ability and AchE of Alzheimer’s Disease Model Rats

摘要 目的:探讨地黄饮子对AD鼠学习记忆及胆碱能损害的影响。方法:模型组以D-半乳糖(D-gal)腹腔注射合并β淀粉样蛋白(Aβ)注射海马制备AD动物模型,各治疗组分别在造模的同时灌胃地黄饮子和抗脑衰胶囊、哈伯因。运用穿梭箱检测各组大鼠的学习记忆能力,采用生化法检测各组大鼠脑匀浆AchE,免疫组化方法观察SYN蛋白的变化。结果:中药治疗组均可使各组大鼠潜伏期降低、主动回避反应阳性率明显升高,被动逃避时间显著降低,表明主动、被动反应水平均有所回升,有显著性差异。灌服中药后各治疗组脑组织AchE活性降低,大鼠海马、皮层神经元SYN蛋白表达增加。结论:地黄饮子对痴呆鼠学习记忆能力的提高及改善神经元损伤的作用,可能与地黄饮子改善鼠脑神经元胆碱能损害,来上调海马和皮层神经元SYN蛋白的表达有关。 Objective :To investigate effects of Decoction of Rehmanniae(DR)on cholinergic neuron injury and Synaptophysin( SYN)of Alzheimer' s disease(AD) medal rats. Methods :Rats induced by D-galactose intraperitoneal injection and the hippocampus injected β-amyloid(Aβ) to make AD models. Every group fed with Hup A.Kangnaoshuai and DR. It can be measured the ability of learning memory by shuttle box. The SYN of hippocampus and cortex neurons were observed by electron lens using immunohistochemistry method. Results : DR can decrease the ability of initiative and passive phobic reaction. DR can improve the ability of learning memory. DR can reduce the activity of AchE in the brain. DR can enchance the expression of SYN protein in the hippocampus and cortex neurons. Conclusion : DR can improve the ability of learning and memory and improve nerons injury. The mechanism of action is possibly relevant that DR can decrease the activity of AchE to enchance the expression of SYN protein in the hippocampus and cortex neurons.
