Improvement of Synthesis process for α-lipoic Acid

摘要 目的合成α-硫辛酸并进行工艺改进。方法以6,8-二氯辛酸乙酯为原料,水为溶剂,在相转移催化剂四丁基溴化铵的作用下,与二硫化钠反应得到硫辛酸乙酯,然后水解、酸化“一锅”反应得到α-硫辛酸。结果所得产物化学结构经红外光谱、核磁共振谱及质谱等确证,总收率为60.0%。结论改进的合成工艺简便、节约了生产成本,更易于工业化生产。 OBJECTIVE To improve the synthesis process of α-lipoic acid. METHODS Ethyl lipoate ester was synthesized by the reaction of 6, 8-dichloro ethyl caprylate and sodium disulfide in aqueous solution of tetrabutyl ammonium bromide, and then hydrolyzing and acidifying in one-pot to yield α-lipoic acid. RESULTS The overall yield was 60%. The structure of α-lipoic acid was confirmed by IR, ^1H NMR and MS. CONCLUSION This synthetic technique was simple, economical and suitable for industry.
