摘要
蒲黄经热水提取,脱除蛋白质、乙醇沉淀,Sephadex G50柱层析分离,DEAE-Sephadex A25和DEAE-Cellulose离子交换柱层析分离得3种白色粉末状多糖TAA、TAB、TAC。经凝胶柱层析、醋酸纤维素薄膜电泳证明,三者为均一体。3种多糖经酸全水解、纸层析和气相色谱分析,确定了糖基的组成及摩尔组成比。用核磁共振、甲基化分析、NaIO_4氧化等方法对多糖结构进行了初步探讨。
Three polysaccharides TAA, TAB, TAC have been isolated, purified from the pollen of Ty-pha angustifolia L. by hot water extraction, alcohol precipitation, sevage method, Gel filtration (Sephadex G50), ion-exchange chromatography on DEAE Sephadex A25 and DEAE-cellulose.The structure determination was carried out by paper chromatography of the hydrolysis products, GLC analysis of the alditol acetates, methylation analysis, 1H NMR and 13C NMR spectra. TAA is mainly composed of a-L-arabinofuranose, β-D-galactose and a-D-galacturonic acid. TAC contains the backbone mainly composed of (1→5)-linked-.L-arabinosyl residue.
