摘要 目的:观察骨髓间充质干细胞(mesenchymal stem cells,MSCs)经5-氮杂胞苷(5-azacytidine,5-Aza)诱导的成肌向分化潜能。方法:分离纯化4周龄SD大鼠的MSCs,以5-Aza诱导,通过细胞形态、免疫组织化学染色观察诱导后不同时相点的细胞。结果:经5-Aza诱导后14d,细胞表达结蛋白(desmin)。细胞形态和排列方式发生明显变化,可见有肌管样细胞出现。结论:MSCs经5-Aza诱导后可以向骨骼肌成肌细胞定向分化。 Objective: To examine the potential of bone marrow mesenchymal stem cells (MSCs) induced into skeletal muscle ceils exposed to 5-azacytidine. Methods: 4-week old rats MSCs were separated and purified, and then induced by 5-azacytidine. Morphological change and immunohistochemisty were observed at various intervals. Results: MSCs began to express desmin after induction by 5-azacytidine at 14th day. The morphology and alignment of the cells changed obviously. The myotubule-like cells were observed. Conclusion: Myoblasts (skeletal muscle) could be differentiated from MSCs under the induction of 5-azacytidine.
