摘要
目的探讨压力超负荷大鼠肥厚心肌高能磷酸盐含量及分布变化的特点。方法成年雄性SD大鼠行腹主动脉缩窄术,术后5周及15周观察大鼠血流动力学参数、心室重构指标,密度梯度法提取大鼠心肌线粒体,高效液相色谱法测量心肌组织及心肌线粒体高能磷酸盐含量。结果大鼠腹主动脉缩窄术后5周出现左心室肥厚,术后15周加重并有心功能减退;CAA5周组与CAA15周组心肌组织ATP、ADP、AMP及总腺苷酸池含量均降低,且CAA15周组心肌PCr降低,其总腺苷酸池及ATP含量低于CAA5周组;CAA15周组线粒体内ATP、ADP、AMP及总腺苷酸池含量降低。结论压力超负荷可影响心肌高能磷酸盐的含量和分布,肥厚心肌组织能量消耗增多且能量产生及储备均不足,可能是心肌发生肥厚及心功能异常的机制之一。
Objective To explore the effects of pressure overload on high energy phosphates content and its distribution of left ventricular hypertrophy in rats. Methods Male SD rats were randomized into 4 groups: coarctation of abdominal aorta 5week group( CAA5 group), 15weekgroup (CAA15 group) and sham operation group(SH5, SH15group). The hemodynamics and ventriclar remodeling parameters were measured. Ventricle of heart were removed and mitochondria were isolated by centrifugation. The size of adenine acid pool(ATP, ADP, AMP) and creatine phosphoric acid(PCr) in myocardium and mitochondria were measured by high performance liquid chromatography(HPLC). Results Left ventricular hypertrophy was observed in operative group. Compared to sham operation group, ATP and content of adenine acid pool decreased significantly in myocardium both in CAA5 group and CAA15 group as well as PCr decreased significantly in myocardium in CAA15 group. ATP and content of adenine acid pool in mitochondria decreased significantly in CAA15 group. Conclusion Myocardial high energy phosphates content and its distribution deranged during pressure overload in rat. These findings suggested that imbalance of energy generation and utilization might play a critical role in hypertrophy and disfunction of heart.
