利用固定化酵母细胞转化反式肉桂酸生产L-苯丙氨酸 被引量:8

Biotransformation of Trans cinnamic Acid to L phenylalanine Using Immobilized Whole Rhodotorula rubra Cells

摘要 研究了深红酵母(Rhodotorularubra)的培养基成分、培养固定化及转化条件。实验表明最佳培养基成分(%):葡萄糖05,胰蛋白胨05,酵母膏05,磷酸二氢钾005,LPhe005,pH70,30℃,20L发酵罐中培养15~17h。最佳固定化条件为:用25%卡拉胶包埋18%的湿菌体。最佳转化条件为:10%反式肉桂酸,4mol/L铵离子,pH105,30℃。 The biotransformation of trans cinnamic acid to L phenylalanine has been studied using phenylalanine ammonia lyase(PAL) containing Rhodotorala rubra .The strains were grown and fully induced for PAL in medium containing:0.5% glucose,0.5% tryptone,0.5% yeast extract,0.05% KH 2PO 4,0.05% L Phe,pH7.0.Cultures were grown at 30℃ in 20L fermenter and harvested after 15~17h growth.For immobilized Rhodotorala rubra cells,maximum conversion degrees were observed using in the trans cinnamic acid concentration of 1.0%,4mol/L NH 4OH.The optimum pH and temperaturefor conversion are 10.5 and 30℃.The best immobilized material and method was that 18% wet cell was immobilized in 2.5% κ carrageenin solution.The κ carrageenin immobilized Rhodotorala rubra cells could convert 77.7% trans cinnamic acid to L phenylanlanine.
