羟丙基-β-环糊精对黄芪甲苷包合作用的研究 被引量:7

摘要 目的制备黄芪甲苷-羟丙基-β-环糊精包合物,并研究其包合作用及增溶效果。方法利用水溶液-搅拌法制备黄芪甲苷-羟丙基-β-环糊精包合物,运用旋光度增量法测定包合比;并利用高效液相色谱法测定黄芪甲苷溶解度及溶出度,以考察其增溶作用。结果羟丙基-β-环糊精与黄芪甲苷可形成摩尔比1∶1的包合物,包合后黄芪甲苷的溶解度由原来的22.2μg.m l-1提高到286.8μg.ml-1,溶出度由26.4%提高到71.2%。结论黄芪甲苷-羟丙基-β-环糊精包合物能显著提高药物的溶解度及溶出度。 Objective To prepare Astragaloside Ⅳ - hydroxypropyl - β - cyclodextrin Inclusion complex, as well as to study the inclusion mechanism and the effect as the solubilizer. Methods Astragaloside Ⅳ - HP - β - CD was prepared by solution - agitation technology, and the inclusion ratio was studied by measuring optical activity. At the same time, the solubility and dissolution were measured by HPLC. Results The inclusion ratio of Astragaloside Ⅳ - hydroxypropyl - β - cyclodextrln was 1 : 1. The solubility of Astragaloside Ⅳ increased from 22.2 μg·ml^-1 to 286. 8 μg·ml^-1, and the dissolution increased from 26.4% to 71.2%. Conclusion The solubility and dissolution of Astragaloside Ⅳ can be improved by preparing the inclusion complex with hydroxypropyl - β - cyclodextrin.
