摘要
人重组骨形成蛋白—2(rhBMP-2)主要生物学作用是诱导新骨形成和促进骨缺损的修复。采用小鼠肌袋模型,观察了大肠杆菌表达的rhBMP-2在不同剂量下的异位成骨活性。实验结果表明,rhBMP-2具有较强的骨诱导活性,其生物学作用具有时间依赖性和剂量依赖性。另外还首次报道了将经综合化学处理的小羊骨松质作为rhBMP-2的载体,使两者有机结合,研制成一种新型复合异种骨,并经小鼠异位诱导实验证实,经综合化学处理的骨松质是rhBMP-2的良好载体,能够显著提高rhBMP-2的骨诱导活性。
Human recombinant bone morphogenetic protein 2 (rhBMP 2) can induce new bone formation and be used for the treatment of bone defect. Bioactivity assay in mouse thigh muscle pouch showed that rhBMP 2 induced the proliferation of mesenchymal type cells and the fomation of new bone in the implantation area in a dose and time dependent manner. Comprehensively chemically treated lamb cancellous bone was combined with rhBMP 2 and implanted into the muscle pouch. The composited graft showed a definitive osteoinductive activity.The data indicate that the treated lamb bone can be used as the carrier and framework for rhBMP 2.
