摘要
目的:观察大鼠小肠缺血再灌注后肺、肝、心、肾组织中bax,bcl-2,p53表达的改变,探讨小肠缺血再灌注损伤对多器官所致的次级损伤。方法:实验于2005-04在延安大学医学中心实验室完成。将64只雄性Wistar大鼠随机分为正常对照组、缺血再灌注0、30min,1、2h,1、3、7d组8组,每组8只。除正常对照组外,其他大鼠均阻断肠系膜上动脉,建立小肠缺血再灌注模型,在再灌注后相应时间点处死大鼠,切取肺、肝、心、肾组织,用免疫组织化学SP法观察各组织中bax,bcl-2,p53的表达情况。结果:64只大鼠进入结果分析。与正常对照组比较,再灌注0min:肺、肝、心、肾组织中bax,bcl-2、p53阳性细胞逐渐增多;再灌注30min时,bax,bcl-2,p53阳性细胞升高更为明显,bcl-2阳性细胞率明显高于bax[肺:(78.13±7.04)%,(17.13±4.60)%;肝:(24.75±5.12)%,(11.25±2.55)%;心:(67.88±5.79)%,(53.63±4.69)%;肾:(22.25±4.82)%,(13.38±3.65)%;P均<0.01];但再灌注至2h时各指标明显降低,其后持续升高;再灌注7d时bax阳性细胞率明显高于bcl-2[肺:(94.12±3.18)%,(83.38±3.54)%;肝:(63.25±9.35)%,(41.25±5.17)%;心:(52.38±4.47)%,(31.88±1.96)%;肾:(59.85±8.25)%,(40.75±6.27)%;P<0.05,0.01]。结论:大鼠小肠缺血再灌注后肺、肝、心、肾这些远隔器官组织细胞内bax,bcl-2,p53等凋亡相关基因表达增强,可能导致远隔器官的次级损伤。
AIM: To observe the expression changes of bax, bcl-2, p53 in the lung, liver, heart and kidney of rats with small intestinal ischemia/reperfusion, and try to find out the seconday injuries in many organs induced by small intestinal ischema/reperfusion. METHODS: The experiment was conducted in the central laboratory of Medical Center, Yan'an University in April 2005. Sixty-four male Wistar rats were randomly divided into control group, ischemia following 0, 30 minutes, 1 and 2 hours, 1, 3 and 7 days reperfusion groups with 8 rats in each group. Except the control group, the superior mesenteric artery of all rats was blocked to establish the models of ischemia and reperfusion in small intestine. The rats were killed after reperfusion for corresponding time to take the lung, liver, heart and kidney tissues. The expressions of bax, bclo2 and p53 in those tissues were observed by immunohistochemical SP method. RESULTS: All 64 rats were involved in the result analysis. Compared with the control group, the ratio of positive cells of bax, bcl-2 and p53 began to increase after 0 minute reperfusion; they increased continuously after reperfusion for 30 minutes, and the rate of positive cells of bcl-2 was higher than that of bax [lung: (78.13±7.04)%, (17.13±4.60)%; liver: (24.75±5.12)%, (11.25±2.55)%; heart: (67.88±5.79)%, (53.63±4.69)%; kidney: (22.25±4.82)%, (13.38±3.65)%, P 〈 0.01]; But they decreased apparently at 2 hours, then increased continuously and the rate of bax was higher than that of bcl-2 at 7 day after reperfusion [lung: (94.12±3.18)%, (83.38±3.54)%; liver: (63.25±9.35)%, (41.25±5.17)%; heart: (52.38± 4.47)%, (31.88±1.96)%; kidney: (59.85±8.25)%, (40.75±6.27)%; P 〈 0.05, 0.01]. CONCLUSION: The expressions of the apoptotic related genes such as bax, bcl-2 and p53 are increased in the distant organs including lung, liver, heart and kidney after small intestinal ischemia/reperfusion, which may result in the secondary lesion of distant organs.
