摘要
为了进一步提高牛体外受精(BIVF)胚的发育率,探讨了来自不同大小卵泡、具有不同卵丘细胞层数以及细胞质形态不同的卵母细胞对牛体外受精效果的影响.结果表明:1)分别采自直径大于5mm、2~5mm和小于2mm卵泡的卵母细胞,经体外成熟、体外受精后,卵裂率分别为54.5%(30/55)、64.4%(85/132)和50.4%(63/125).其中,2~5mm卵泡的卵母细胞的体外受精结果显著高于小于2mm卵泡的卵母细胞(P<0.05).三类卵母细胞的囊胚发育率分别为25.4%(14/55)、24.2%(32/132)和16.0%(20/125),三者之间无显著差异;2)根据卵丘细胞层的多少,将卵母细胞分为带有5层以上致密卵丘细胞、3~5层较疏松卵丘细胞和1~2层疏松卵丘细胞或半裸卵三类.此三种卵经体外受精以后的卵裂率分别为:60.0%(90/150)、69.0%(100/145)和45.8%(49/107);囊胚发育率分别为20.0%(30/150)、21.4%(31/145)和5.6%(6/107).前两种卵母细胞的卵裂率和囊胚发育率显著高于第三种(P<0.05,P<0.01);3)根据细胞质形态,将卵母细胞分为?
In order to enhance development rate of bovine in vitro fertilization (BIVF) embryo,these experiments were designed to investigate the effects of oocytes from different follicle size oocytes with different cumulus cells and oocytes with different ooplastic morphology on IVF.The results showed that 1)The cleavage rates of oocytes aspirated from >5 mm、2 ̄5 mm and <2 mm follicle in diameter were 54.5%(30/55)、64.4%(85/132) and 50.4%(63/125) b (a>b, P <0.05) respectively,after in vitro maturation and fertilization, and the development rates of blastocysts were 25.4%(14/55)、24.2%(32/132) and 16.0%(20/125),respectively;2) The oocytes were classified into three groups according to the layers of cumulus cells:A type oocytes are those with more than 5 layers of compact cumulus cells、B type oocytes are those with 3 ̄5 layers of cumulus cells and C type oocytes are those with 1-2 layers of cumulus cells or partly denuded oocyte,respectively.The cleavage rates of above 3 types oocytes were 60.0%(90/150) a、69.0%(100/145) b and 45.8%(49/107) c after in vitro maturation and fertilization,and the development rates of blastocysts were 20.0%(30/150) a、21.4%(31/145) b and 5.6%(6/107) c (a>c, P <0.05;b>c, P <0.01);3) The oocytes were graded into 2 categories on ooplasmic characteristics:Category 1,with dark ring in the periphery of the plasm or with heterogeneous ooplasm;Category 2,with homogeneous transparent ooplasm.The cleavage rate and blastocysts development rate of category 1 oocytes after in vitro maturation and fertilization were 73.5%(72/98) and 22.4%(22/98),respectively,they were significantly higher than that of category 2 oocytes (54.0%,40/74, P <0.01; 10.8% ,8/74, P <0.05).
