半枝莲对人大肠癌细胞增殖抑制及诱导凋亡的作用 被引量:24

The Effect of Scutellaria Barbata D. Don on Inhibiting the Proliferation of Human Colorectal Cancer Cell Line HT-29 and Inducing Its Apoptosis

摘要 目的探讨半枝莲(SBD)在体外对人大肠癌HT-29细胞增殖抑制、诱导凋亡作用及可能的作用机制。方法应用MTT法(噻唑蓝比色试验)检测不同浓度的SBD对体外培养的人大肠癌HT-29细胞增殖的抑制作用,流式细胞仪技术测定SBD对HT-29细胞凋亡及细胞周期分布的影响。结果HT-29细胞经SBD(浓度范围10~320 mg/L)作用后细胞生长明显受到抑制,呈现明显的剂量依赖效应关系和时间依赖效应关系,最高抑制率为93.869%(P〈0.01);SBD在20mg/L、80mg/L、320mg/L 3种浓度下作用48h均可诱导HT-29细胞凋亡,流式细胞仪技术检测其凋亡率分别为7.6%、16.2%、和34.5%,有明显的剂量依赖效应关系;同时,SBD使细胞周期分布发生明显变化,表现为S期细胞比例上升,G0/G1期和G2/M期细胞比例下降。结论SBD能抑制HT-29细胞增殖,而且能诱导HT-29细胞凋亡,其作用机制可能与影响癌细胞细胞周期的分布有关。 Objective To investigate the effects of scutellaria barbata D. Don(SBD) in inhibiting proliferation of HT-29 cells of human colorectal cancer cell line and inducing its apoptosis and the possible molecude mechanism . Methods The inhibiting effect of SBD with different concentration on human colorectal cancer cell line in vitro were detected by MTF assay. The induction of apoptosis and the influence on the distribution of HT- 29 cell cycle were measured by flow cytometry. Results SBD at the concentration of (10 -320)mg/L could inhibit the proliferation of HT-29 cells , which showed obvious concentration-effect and time-effect relationship, and the hightest inhibition rate was 93. 869% ( P 〈 0.01 ) ; SBD at the concentration of 20mg/L, 80mg/L, 320mg/L could induce apoptosis of HT-29 cells, the rates were 7.6%, 16.2% and 34.5%, respectively, which showed obvious concentration-effect relationship. The cell cycle distribution was altered simultaneously, sphase of the cells were increased , while G0/Gl and G2/M phase of the cells were decreased after treated by SBD. Conclusion SBD may inhibit proliferation of HT-29 cells, and could induce apoptosis of HT-29 cells. The possible mechanism is the influence in distribution of cancer cell cycle.
