摘要
用作者改进的液体培养基在5%CO_2环境中培养霍乱弧菌(Inaba 569B),每ml培养滤液可获得霍乱肠毒素(CT)1.215~2.012μg。在分离提纯CT时,先用DEAE-Sephadex A_(50)除去大部分色素及杂蛋白,再用Sephacryl S_(-200)分纯,效果好,重复性强;其第Ⅰ组分即为CT,肠襻试验阳性,双向琼脂扩散试验及HPLC检测结果均与上海CT一致。分子量为86000。
The vibrio cholera (Inaba 569B ) being cultured with our improved liquid medium in the environment of CO2, the cholera enterotoxin (CT ) may be obtained about 1.215-2.012ug/ml culture filtrate.When the CT was isolated and purified from the culture filtrate, the DEAE- Sephadex A50 was first used to remove the most of the pigment and the other mixed proteins, and then the Sephacryl S-200 was done with a good result and great repeatability.The firstfract ion eluted from Sephacryl S -200 column was CT , its ligatured small intestine segment test was positive, the results of double diffusion test and HPLC detection of the first fraction were all the same as the Shanghai CT, and its siolecular weight was 86 000.
