重组hEGF-TCS融合蛋白基因表达载体的构建及表达产物的纯化 被引量:1

摘要 目的构建新型人表皮生长因子(human epidermal growth factor,hEGF)和天花粉蛋白(trichosanthin,TCS)融合蛋白的表达载体,在大肠杆菌中表达并纯化EGF-TCS融合蛋白。方法利用基因工程技术将人表皮生长因子EGF和天花粉蛋白基因连接起来,克隆到高效表达载体pQE30中,构建重组表达质粒pQE30-EGF-TCS。并在大肠杆菌M15中表达该融合蛋白(EGF-TCS)。产物经Ni-NTA Agrose亲和层析柱纯化。结果重组表达质粒pQE30-EGF-TCS在大肠杆菌M15中获得稳定、高效的融合表达。目的蛋白占细胞总蛋白的40%左右,以可溶性形式存在,表达上清柱纯化纯度达95%以上。结论应用基因工程技术,成功构建了融合蛋白EGF-TCS,为进一步研究其结构功能关系和临床应用奠定了基础。
