扁藻多糖的分离分析及其生物活性的初步研究 被引量:9

Separation and Analysis of Polysaccharide from P. subcordigoramis (Will) Hanzen and Primary Studies on It’s Anticancer Activity

摘要 目的:初步确定亚心型扁藻多糖的理化性质、结构特征及抗肿瘤活性。方法:从培养至平衡期后的亚心型扁藻细胞内分离出乙醇沉淀的多糖级分,经过Sephadex G-150凝胶层析纯化后,利用化学分析及仪器分析进行理化性质和结构特征分析,并通过此多糖对小鼠体内黑色素瘤的抑制作用确定其抗肿瘤活性。结果:扁藻多糖含有氨基和硫酸基,并且含有以葡萄糖为主的多种已知和未知的单糖糖基和糖醛酸,其对小鼠体内黑色素瘤的抑制率为34.2%。结论:此实验结果为扁藻多糖在医疗方面作为抗肿瘤药物的可能性提供了基础依据。 Objective: To determine the physical and chemical properties of polysaccharide from Platymonas subcordigoramis ( Will ) Hanzen and it's structural character and anticancer activity. Methods: The polysaccharide was separated by ethmlol - precipitation from Platymonas subcor- digoramis (grdl) Hanzen grown in the stationary phase and was purified through SephadexG- 150 gel chromatography, it's physical and chemical properties and structural character were analyzed by means of chemical analysis and instrumental analysis, and the anticancer activity was determined by inhibiting the growth of melanoma in mice. Results: The polysaccharide from P. subcordigoramis (Will) Ho.nzen contained- NH2 and -SO4, and comprised many kinds of known and unknown monosaccharide groups and alduronic acids, among of the known monosaccharide groups, glucose group was on the principal part. The irthibition rote of melanoma in mice was 34.2%. Conclusion: The results provided the probability for the pelysaccharide from P. subcordigoramis ( Will ) Hanzen as an anticancer drug in medical field after experiencing an embedded, systemic and long time research.
