摘要
研究了菊粉酶高产菌株-黑曲霉M89的产酶条件,产酶的最适碳源是菊粉,最适氮源是草酸胺、硫酸铵和磷酸二氢铵;250mL摇瓶装50mL培养基,在培养基起始pH7.0,32℃,210r/min条件下摇振培养产酶活力最高。黑曲霉M89菊粉酶的最适反应温度为55~60℃,最适PH4.5,在60℃热处理3h,仅失活17%,在PH4.0~9.0范围内室温处理12h,无活力丧失。
Inulinase yielding related factors of strain Aspergillus niger M,, were studied.The strain gave the highest enzyme levels with inulin as catbon source. The optimalnitrogen substrates for inulinase production of the strain were ammonium oxalate,(NH4)2SO4 and NH4H2PO4. Aspergillus niger M,,produced maximal inulinase activity withthe medium initial pH7. 0 at temperature 32℃ 210r/min Stirring culture. The optimaltemperatrue for the inulinase activity was between 55~60℃ with pH4. 5. Incubated underthe condition of 60℃ for 3h,the inulinase activity Iosed only 17%. At room temperature,dialyzed with pH4. 0~9.0 buffer for 12h,Inuliuase activity could be recovered by 100%'
