摘要
目的:建立一种白假丝酵母菌培养和鉴定的方法。方法:选用经改良沙氏培养基培养白假丝酵母菌,观察假菌丝、厚膜孢子,芽管形成,并结合糖发酵和其他生化试验给以初步鉴定,新法提取DNA作最后鉴定。结果:形态学观察和生化反应证实菌种为Candida albicans1型。DNA提取表明菌种与标准株DNA带型完全相同,提取DNA浓度(1.06±0.29)μg/μl,纯度好(OD260/OD280>1.7)。结论:本法制备的培养基适合白假丝酵母菌的生长和鉴定。
To find out a method of isolating and identifying Candida albicans . Methods ： Candida albicans were selected and cultured by improved Sabourand＇ s medium for observing pseudodypha, chlamydospore and the germ tube formation. The strain was initially identified by the combination of sugar fermentation and other biochemical tests, and by a new method of extracting DNA, Candida albicans were identified finally. Results： Morphological and biochemical tests verified that the strain was type Candida albicans 1, DNA extraction showed belt type was identical with the standard strain and concentration of DNA extraction was （ 1.06 ± 0.29 ） μg/μl with high purity （OD260/OD280 〉 1.7）. Conclusion ： The medium cultured by this method can fit for the growth and identification of Candida albicans.
