摘要
目的:建立一种白假丝酵母菌培养和鉴定的方法。方法:选用经改良沙氏培养基培养白假丝酵母菌,观察假菌丝、厚膜孢子,芽管形成,并结合糖发酵和其他生化试验给以初步鉴定,新法提取DNA作最后鉴定。结果:形态学观察和生化反应证实菌种为Candida albicans1型。DNA提取表明菌种与标准株DNA带型完全相同,提取DNA浓度(1.06±0.29)μg/μl,纯度好(OD260/OD280>1.7)。结论:本法制备的培养基适合白假丝酵母菌的生长和鉴定。
To find out a method of isolating and identifying Candida albicans . Methods : Candida albicans were selected and cultured by improved Sabourand' s medium for observing pseudodypha, chlamydospore and the germ tube formation. The strain was initially identified by the combination of sugar fermentation and other biochemical tests, and by a new method of extracting DNA, Candida albicans were identified finally. Results: Morphological and biochemical tests verified that the strain was type Candida albicans 1, DNA extraction showed belt type was identical with the standard strain and concentration of DNA extraction was ( 1.06 ± 0.29 ) μg/μl with high purity (OD260/OD280 〉 1.7). Conclusion : The medium cultured by this method can fit for the growth and identification of Candida albicans.
