测序法及液相芯片杂交法对生殖道人乳头瘤病毒感染分型的比较 被引量:8

摘要 目的探讨人乳头瘤病毒(HPV)分型方法在临床标本应用中的特异性及敏感性。方法采用通用型引物用PCR方法对尖锐湿疣及宫颈细胞内瘤变组织标本进行HPV DNA检测,测序法及液相芯片杂交法(luminex法)对其进行分型。结果在单型感染中测序法较为准确,而在多型感染中luminex法占一定优势。结论测序法结合luminex法可以运用在HPV感染标本中的分型。 Objective To compare the specificity and sensitivity of two genotyping approaches for human papillomavirus (HPV). Method HPV DNA was amplified and detected in ciinical specimens by poly- merase chain reaction in a pair of universal primers MY09/11, and then genotyped with either sequencing method or liquid chip hybridization method ( luminex method). Result Sequencing method obtained precise genotyping results in single-type HPV infection, while luminex method obtained accurate genotyping results in multiple-type HPV infection. Conclusion A combined method using both sequencing and luminex method is suitable for the genotyping of HPV-infected specimens.
