乙型肝炎患者HBcAg18-27表位特异性细胞毒性T细胞的研究 被引量:19

摘要 目的对乙肝患者尤其是慢性乙肝患者外周血HBcAg18-27表位特异性细胞毒性T细胞(CTL)的分布频率及功能状态进行研究。方法应用多色流式分析结合HLA-A*0201限制性表位肽/五聚体复合物技术对乙肝患者外周血HBcAg18-27表位特异性CTL进行精确定量,应用胞内细胞因子染色技术结合流式细胞分析技术研究HBcAg18-27表位特异性CTL穿孔素,颗粒酶B,干扰素-γ的分泌水平。结果在急性与慢性乙型肝炎患者外周血中均可测到HBcAg18-27表位特异性CTL,两者的分布频率存在显著性差异。慢性乙肝患者外周血中HBcAg18-27表位特异性CTL分泌穿孔素,颗粒酶B,干扰素的水平明显降低。结论HBcAg18-27表位特异性CTL在乙型肝炎病毒清除中起到重要作用,慢性乙肝患者外周血HBcAg18-27表位特异性CTL存在功能缺陷。 Objective To investigate the frequency and function of HBcAg18-27 specific cytotoxic lymphocytes (CILs) in the patients with hepatitis B virus infection. Methods The frequency of HBcAg18-27 specific CTLs was quantified by poly-cytometry and HLA-A * 0201 limited epitope-peptide/pentamer assy. The secretions of perforin, grazyme B, and IFN-γ from HBcAg18-27 specific CTLs were analyzed by ICC.Results The frequency of HBcAg18-27 CFLs was significantly difference between the patients with acute and chronic hepatitis B virus infection. While the secretions of perforin, grazyme B, and IFN-γ were reduced in patient with chronic hepatitis B virus infection. The HBcAg18-27 specific CILs may play a crucial role in the clearance of hepatitis B virus. There is a function defect of HBcAg18-27 specific CTL in CHB patients.
