高渗盐溶液对失血性休克大鼠细胞免疫及p38MAPK信号通路的影响 被引量:3

摘要 目的探讨7·5%氯化钠高渗盐溶液(HSS)对失血性休克大鼠脾脏T淋巴细胞功能的影响以及p38MAPK信号转导途径是否参与该过程。方法SD雄性大鼠遭受失血性休克(平均动脉压40mmHg)60min后分别用生理盐水、HSS以及HSS+SB203580(SB)10mg/kg(ip)复苏。免疫荧光观察脾T淋巴细胞中p38蛋白的表达水平,同时ELISA法检测脾T淋巴细胞IL-2含量,MTT法测定脾T淋巴细胞增殖率。结果免疫荧光显示HSS组T淋巴细胞p38MAPK表达显著高于NS组和HSS+SB组,HSS组脾淋巴细胞增值率显著高于NS组和HSS+SB组(P<0·01),HSS组IL-2含量显著高于NS组和HS+SB组(P<0·01)。结论HSS可能通过p38MAPK信号转导途径改善失血性休克机体的脾脏T淋巴细胞功能。 Objective To evaluate the effects of resuscitation with hypertonic saline solution (HSS) on the splenic T lymhocytes in rats with hemorrhagic shock. Methods Controlled hemorrhagic shock was induced by blood withdrawal (MAP, 40 mmHg) and maintained for 60 min. Animals were randomly divided into 3 groups. Group 1 (n= 10) was treated with 30 ml/kg normal saline(NS), group 2 (n=10) with 4 ml/kg HSS and SB203580 (1 mg/kg,ip), and group 3 (n=10) with 4 ml/kg HSS. The rats were killed 24 h after resuscitation. Spleen T lymphocytes were separated, the expression of p38MAPK protein in the lymphocytes was determined by immunol fluorence,and MTT and ELISA were used to determine the IL-2 of T lymhocytes and the ability of splenic lymhocytes proliferation. Results The expression of p38MAPK protein in the spleen lymphocytes was increased significantiy in group 3 than that in group 1 and group 2. The productive content of IL-2 in group 3 was significantly higher than that in group 1 and group 2. The T lymhocytes proliferation rate in group 3 was significantly higher than that in the other two groups. Conclusion HSS may improve the function of splenic T lymhocytes in hemorrhagic shock rats through the p38MAPK pathway.
