摘要
目的:探讨乙酰肝素酶(heparanase,HPA)及缺氧诱导因子-1α(hypoxia inducible factors-1α,HIF-1α)在食管鳞癌中的表达及其与食管鳞癌浸润转移的关系。方法:应用原位杂交技术检测70例食管鳞癌组织和23例切缘正常组织中HPA mRNA表达,同时应用免疫组化法检测HIF-1α蛋白在全部标本中的表达。结果:癌组织中HPA mRNA和HIF-1α的阳性表达率分别为65.7%(46/70)和55.7%(39/70),显著高于切缘正常组织(P<0.05);HPA mRNA和HIF-1α的表达与食管鳞癌组织浸润深度、淋巴结转移有关(P<0.05);在食管鳞癌组织中HPA mRNA的表达与HIF-1α的表达关系密切(Kappa=0.615,P<0.05)。结论:HPA和HIF-1α阳性表达与食管鳞癌的生长及浸润转移有关,两者在食管鳞癌的发生、发展过程中可能存在着一定的协同作用;检测HPA与HIF-1α表达有助于食管鳞癌生物学性能的判断。
Objective: To investigate the relationship of expression of HPA and HIF-1α in human esophageal squamous cell carcinoma with its invasion and metastasis. Methods:The expression of HPA mRNA and HIF-1α protein in 23 normal tissue specimens of incised margin and 70 esophageal squamous cell carcinoma specimens were examined by in situ hybridization assay and immunohistochemical stain respectively. Results: HPA mRNA and HIF-1α protein were positive in 65.7% and 55.7% of all 70 cancer tissue specimens, respectively,which were significantly higher than those in incised margin normal tissues(P 〈0.05). The high expression of HPA mRNA and HIF-1α protein in the esophageal squamous cell carcinoma was significantly correlated with fibromembranous infiltration and lymph node metastasis( P 〈0.05 ). There was a close correlation between the expression of HPA mRNA and HIF-1α protein in the esophageal squamous carcinoma tissues (Kappa = 0. 615 ,P 〈 0.05 ). Conclusion: The high expression of HPA mRNA and HIF-1α protein is correlated with carcinogenesis progression,invasion and metastasis of esophageal squamous cell carcinoma. There is a synergic action between them in the carcinogenesis and development of esophageal carcinoma. The determination of HPA and HIF-1α will be useful for predicting tumor biological behavior.
