摘要
目的:直径1.0~1.4mm静脉经二步冷冻法保存(第一步-60℃,第二步-196℃)与异体移植,观察维持时限对二步法静脉冷冻保存的影响。方法:家兔60只,体重2.0~2.5kg,切取股静脉段(20~25mm),分成6组,每组10只;对照组为自体静脉移植,余5组在-60℃中按维持时限5min、15min、30min、60min、120min分组。用慢降温0.5~1.0℃/min,快复温后作异体静脉移植。术后经光镜、电镜、血管造影、手术探查等对移植血管的组织学及通畅率检测,采用四格表确切概率法检验。结果:60min组内膜保存最好,远期(12周)通畅率89.9%与对照组差异无显著意义(P>0.05);其余各组静脉内膜损伤严重,通畅率与对照组差异显著(P<0.05)。结论:(1)静脉冷冻保存维持时限有一定选择性;(2)60min组为最佳维持时限;(3)为临床应用保存异体小静脉修复小血管缺损提供依据。
Objective:Veins(Φ1.0~1.4 mm) were cryopreserved in a two step procedure and transplanted allogenously in order to study the role of holding time in venous cryopreservation.Methods:Femoral veins taken from rabbits,were preserved in two step procedure:holding first at -60℃ for various times (5,15,30,60,120 min) and then shifted into -196℃ in liquid nitrogen for 48 hours.After thawing,part of these cryopreserved veins was used for light and electron microscopic examination and part for allograft with fresh autograft as control,and angiography was performed 1 wk after operation.Results:Veins were shown to be best preserved in the holding time of 60 min.The patency rate was 89.9% in 12 wk after operation with no significant difference from that of the fresh autograft( P ≥0.50),whereas,the veins cryopreserved in other holding times,the damage was severe.The patency rate differed greatly from that of the control( P <0.05).Conclusion:These results indicated that the holding time play an important role in vein cryopreservation,the optimal holding time was 60 min.
