摘要
目的探讨益母草生物碱对大鼠心肌梗死后心功能保护作用的机制。方法采用左冠状动脉前降支结扎制作大鼠心肌梗死模型,通过大鼠血流动力学指标以及心肌肌浆网钙泵的活性(SERCA2α)及其蛋白水平来反映益母草生物碱对大鼠心肌梗死后心功能的影响不明显(P<0.05)。结果益母草生物碱可以显著提高模型大鼠左室收缩压以及左室内压最大上升速率(P<0.05),降低模型大鼠左室舒张末压以及明显提高左室内压最大下降速率(P<0.05);益母草生物碱对模型大鼠心肌组织SERCA2a活性及SERCA2a含量均有显著提高(P<0.05)。结论益母草生物碱能够提高冠状动脉结扎大鼠心肌收缩舒张功能,其可能机制是提高大鼠心肌组织SERCA2a活性及含量。
Objeetive To investigate the protecting mechanism of Alkaloid of leonurus on heart function in rats with myocardial infarction (MI). Methods Three days later after left coronary artery ligation, the survival rats were randomly divided into model group, carvedilol group [30mg/(kg·day)] and Alkaloid group[30mg/(kg·day)]. Normal group and sham group were set up too. After four weeks of administration, hemodynamic parameters were measured by inserting catheters and SERCA2α activities and SERCA2α protein of the myocardium were examined. Results There were significant effects on elevating the maximal left ventricular systolic pressure and maximal rate of rise of left ventricular pressure in the group of alkaloid (P 〈 0.05). The maximal rate of fall of left ventricular pressure raised obviously but the left ventricular end diastolic pressure (LVEDP) not changed evidently in comparison with model group. The SERCA2α activities and the contents of myocardial SERCA2α were raised in the group of alkaloid (P〈0.05) compared with that of the model group. Conclusion Alkaloid could promote the contraction function of myocardium in rats after coronary artery ligation. The mechanism might he that the Alkaloid can elevate the SERCA2α'contents and activities of myocardium in rats.
