摘要
目的观察黄芪甲苷对大剂量异丙肾上腺素造成的大鼠心肌功能与组织损伤的作用。方法18只SD大鼠分为三组。正常对照组(n=5):腹腔注射0.1 mL.kg-1.d-1黄芪甲苷溶剂,连续3周;异丙肾上腺素对照组(n=8):大剂量异丙肾上腺素(10 mg.kg-1.d-1)皮下注射,连续2 d,并注射黄芪甲苷溶剂(同正常对照组);黄芪甲苷保护组(n=5):腹腔注射黄芪甲苷溶液(5 mg.kg-1.d-1)3周,并于开始注射2 d后皮下注射大剂量异丙肾上腺素(10 mg.kg-1.d-1),连续2 d。给药3周后,各组动物分别进行左室心功能和心肌病理组织学检查。结果大剂量异丙肾上腺素注射能显著降低左室压最大上升/下降速率(dp/dtmax)达22%(P<0.01),并造成显著的心肌损伤。黄芪甲苷能使dp/dtmax和dp/dtmin分别增加94%和84%(P均<0.01),收缩压上升10%(P<0.05),有效增强心功能,减少异丙肾上腺素所致的心肌损伤和纤维化。结论黄芪甲苷可预防大剂量异丙肾上腺素造成的心脏损害。
Objective To observe the effect of astragaloside Ⅳ on isoproterenol-induced myocardial injury. Methods Eighteen SD rats were divided into three groups. Normal control group ( n = 5) :intraperitoneal injection of sdvent ( 0.1 mL· kg^- 1· d^ - 1 ) for three consecutive weeks ; isoproterenol control group( n = 8) :subcutaneous injection of large dose of isoproterenol( 10 mL· kg^- 1· d^ - 1) for two consecutive days, and intraperitoheal injection of sdvent (0.1 mL· kg^- 1· d^ - 1 ) for three consecutive weeks ; astragaloside protection group (n = 5): intraperitoneal injection of astragaloside Ⅳ (5 mg·kg^-1 ·d^-1) for three consecutive weeks, and subcutaneous injection of large dose of isoproterenol ( 10 mL· kg^- 1· d^ - 1 ) for two consecutive days just from the beginning of astragaloside Ⅳ injection. Three weeks after administration, eardial function of left ventricle was measured and myocardial pathological examination was performed. Results Injection of large dose of isoproterenol significantly decreased dp/dtmax of the left ventricle by 22% (P 〈 0.01 ) and caused myocardial injury. However, astragaloside Ⅳ injection increased dp/dtmax, and dp/dtmix by 94% and 84% , respectively( both P 〈0.01 ) , systolic pressure by 10% (P 〈 0.05), and effectively enhanced the eardial function and attenuated the myocardial injury and fibrosis. Conclusion Astragaloside has protective effect on myocardial injury caused by large dose of isoproterenol.
