乙酰甲胺磷对雄性大鼠生殖细胞及睾丸标志酶活性的影响 被引量:4

摘要 目的研究乙酰甲胺磷对雄性大鼠生殖细胞及睾丸标志酶活性的影响。方法30只健康成年SD雄性大鼠,随机分为5组,每组6只。实验组设3个不同剂量乙酰甲胺磷染毒组、阴性对照组(蒸馏水)和阳性对照组(环磷酰胺,20mg/kg)。染毒组和阴性对照组采用经口灌胃染毒方式,阳性对照组采用腹腔注射染毒方式。检测精子数量、精子活率、精子畸形率及睾丸标志酶LDH、SDH、ATPase(Na+K+-ATPase、Mg2+-ATPase、Ca2+-ATPase)的活性。结果乙酰甲胺磷染毒组精子数量、活率下降,精子畸形率升高,睾丸标志酶LDH、SDH、Na+K+-ATPase、Mg2+-ATPase、Ca2+-ATPase等的活性降低,与阴性对照组相比差异有统计学意义(P<0.05);且精子畸形率与染毒剂量呈正相关,而精子数量、精子活率及睾丸标志酶(LDH、SDH、Na+K+-ATPase、Mg2+-ATPase、Ca2+-ATPase等)的活性与染毒剂量呈负相关。结论乙酰甲胺磷可引起雄性大鼠精子数量和活率的降低、精子畸形率的增加及睾丸标志酶活性的降低,提示乙酰甲胺磷具有一定的生殖毒性。 Objectives To explore the effects of acephate on male rat's sperms and the activity of testicular marked enzymes. Methods Different dosage of acephate(0, 11.81, 23.63, 47.25 mg/kg· bw) were orally administered to the adult male SD rats for 60 days. The sperm count, the rate of sperm mobility and aberration, the level of lactate dehydrogenase (LDH), sorbitol dehydrogenase (SDH), ATPase (Na^+ K^+-ATPase, Mg^2+- ATPase, Ca^2 + -ATPase) were detemined. Results The sperm count, the rate of sperm mobility and the level of LDH, SDH, Na^+ K^+ -ATPase, Mg^2+ -ATPase, Ca^2+ -ATPase of the acephate groups were lower, and the rate of aberration was upper. The differences between two groups were significant ( P 〈 0.05). And all the index showed clear and definite dose-effect relationship. Conclusions Acephate has obvious reproductive toxicity on male rats. It can reduce their sperm count and the rate of sperm mobility, increase the rate of aberration and inhibit the activity of testicular marked enzymes.
