摘要
采用寇氏法进行中药复方提取液的急性毒性试验和用药物累积法进行其在鲫鱼体内的药物动力学研究。结果表明复方提取液的24 hLC50=1.207 g/mL,48 hLC50=1.101 g/mL,安全剂量0.785 g/kg。表观药动学参数:零时药物浓度C0=1.4817 g/kg,一级消除速率常数Kel=0.1063 h-1,消除半衰期t1/2=6.0113 h,清除率CLb=0.5851(g.h)/kg,药-时曲线下面积AUC=13.9388(g.h)/kg,表观分布容积V=1.9262。说明该复方提取液在鲫鱼体内代谢较快,药物的生物利用度高,药物与血浆蛋白结合率低,主要分布在组织中。
This paper the acute toxicity of one compound Chinese herbal medicine and the pharmacokinetics of this medicine in Carassius auratus by drug cumulative method. The results are 24 h LC50 = 1.207g/mL, 48 h LC50= 1.101 g/mL, safe dose: 0.785 g/kg. The apparent pharmacokinetics parameters are C0= 14.817 g/kg, Ke1=0.1063 h^-1, t1/2=6.0113 h, CLb =0.5851 (g·h)/kg, AUC = 13.9388 (g·h)/kg, V= 1.9262 which shows that the metabolism of this medicine in carassias auratus is fast, that the medicine can be utilized biologically and it is mostly distributed in tissue.
