摘要
采用PCR法从精选白皮苦瓜(JX)、穗农牌早优苦瓜(ZY)、碧翠苦瓜(BC)、银田牌长白苦瓜(YT)和银田牌蓝山大白苦瓜(LAN)5个品种中克隆到5个长度均为861bp的MAP30基因JX—MAP30、ZY—MAP30、BC-MAP30(Genebank accession No:DQ643967)、YT—MAP30(No:DQ643968)和LAN-MAP03。序列分析表明,BC-MAP30和YT-MAP30与已报道的MAP30基因分别存在2个和3个碱基的不同,均编码263个氨基酸残基组成的成熟蛋白,与其他葫芦科植物的Ⅰ型核糖体失活蛋白类以及天花粉毒蛋白、多花白树毒蛋白、肥皂草毒素的同源性分别为68.90%、82.75%、71.25%和66.45%,其90~220位氨基酸序列较为保守。二级结构预测表明,BC—MAP30和YT—MAP30编码的成熟肚在N端均有一个延伸的6个β片层。
In this study, five MAP30 (momordica anti-HIV protein of 30 kD) genes (JX-MAP30, ZY-MAP30, BC - MAP30, YT - MAP30 and LAN - MAP30) were cloned from five Balsam pear (Momordica charantia) cultivars. 861 bp sequence was obtained by using polymerase chain reaction (PCR). Sequence analysis revealed that there were two different bases in BC- MAP30 and three bases in YT - MAP30. The open reading frame of the gene encoded a 263 - amino acid matured peptide. The sequence identity with other cucurbitaceous Ribosome inactivating proteins (RIPs) was up to 68.90%, with TCS was 71.25%, with Gelonin was 71.25%, and with Saporin was 66.45%. The amino sequence 90 to 220 was conservative sequence. Secondary structure showed that matured peptides encoded by BC- MAP30 and YT - MAP30 formed six successive β sheet.
