减压对慢性压迫性脊髓损伤大鼠凋亡基因bax和bcl-2表达的影响 被引量:1

INFLUENCE OF DECOMPRESSION ON THE BEHAVIORAL FUNCTION AND EXPRESSION OF BAX AND BCL-2 IN CHRONIC SPINAL CORD INJURY OF RATS

摘要 目的探讨手术减压治疗慢性脊髓损伤的机理。方法以45只2个月龄的Wistar大鼠为受试对象,采用大鼠后路渐进性脊髓压迫模型并给予手术减压,然后进行联合行为评分(CBS)、常规病理和免疫组化染色,用原位末端标记法(TUENL)检测凋亡基因bcl-2和bax的表达。结果减压组中bax蛋白表达较压迫组明显减少,凋亡指数(AI)低;而bcl-2表达则较压迫组多,着色深,且CBS降低(p<0.05)。结论手术减压治疗慢性脊髓损伤可能是通过调控bax和bcl-2的表达,从而影响实验动物的行为功能。 Objective To study the mechanisms of the decompression on chronic spinal cord injury(SCI) of rats. Methods According to the chronic progressive compressive spinal cord injury model of rats. 45 wistar rats were divided into 3 groups:Control group, SCI group and decompression group. Each group had 15 rats respectively. Hematoxylinosin staining,immunohistoehemistry and TUENL methods were used to examine the expression of baxand bcl - 2 in rats, spinal cord. The positive immunohistochemistry cells of bcl - 2 and bax were observed by light microscope and analyzed by computer image analysis system. Results After decompression,CBS was decreased and the expression sequences of bcl - 2 protein were observed more obvious than those in SCI group. On the contrary, the bax expresses obviously in the SCI group than those in the decompression group. Conclusion The decompression may increase synthesis of bcl - 2 in spinal cord and facilitate the recovery of behavioral function of experimental animals.
