激素、蛋白质类物质的^(125)I标记率及标记产品的脱碘率测定

摘要 采用氯胺-T法对胰岛素、牛血清白蛋白和猪超氧化物歧化酶进行125I标记,标记率采用淋洗分离时标记产品峰计数率与诸峰计数率之和的比值、三氯醋酸(TCA)沉淀、纸层析及纸电泳等四种方法进行测定。测定结果表明,第一种方法结果偏低,与后三种方法有显著差异,而后三种方法之间无显著差异,其中尤以TCA沉淀法操作简单、方便、速度快。还采用TCA沉淀法对上述几种125I标记产品的保存条件作了探讨,结果提示,标记产品加蛋白保护,并且在低温条件下或以冻干品形式存放时脱碘率最低。 Chloramine-T was used for 125 I labelling of insulin,bovine serum albumin and pig superoxide dismutase.The labelling rates were measured by four methods,i.e.comparing the counts from the labelled compound with those from the eluate,TCA precipitation,paper chromatograph and paper electrophoresis.It was shown that the labelling rate obtained with the first method was the lowest but those with other three methods weres imilar.Especially the TCA precipitation was considered to be the best because of its easiness and rapidit.In addition,the deiodated rates of the labelled compounds in various conditions were measured by TCA precipitation,and the lowest deiodated rate was observed for the products with some protein protection and stored at the temperature lower than -20℃ or as a lophilized state.
