摘要
应用玫瑰花环法分离人外周血T、B淋巴细胞,T_4McAb标记淋巴细胞后经细胞亲和层析分离得到T_4^+和非T_4^+细胞。部分T、B、T_4^+和非T_4^+细胞接受10 Gy^(60)Coγ线照射。将各种细胞匹配分组培养,以~3H-TdR参入法观察PWM对淋巴细胞及其亚群激活功能和^(60)Coγ线的辐射效应。结果表明T、B淋巴细胞均可被PWM激活,B强于T细胞;PWM对B和非T_4^+细胞激活作用相当,唯T_4^+细胞最弱。PWM激活时。T与B、T_4^+与B细胞其DNA合成均有协同作用,前者的协同作用更大。当T、T_4^+或B细胞受到10Gy^(60)Coγ线照射后,它们之间的协同作用消失。非T_4^+与B细胞间既无协同作用亦无抑制作用。10 Gy^(60)Coγ线照射后,B和非T_4^+细胞~3H-TdR参入下降比T_4^+细胞严重,前二者的差别不显著。
T and B lymphocytes in human periphery blood were seperated byrosetting procedure. After lymphocytes were labelled with T_4^+ McAB, T_4^+ and non-T_4^+lymphocytes were isolated by cell affinity chromatography method. When parts of T,B. T_4^+ and non-T_4^+ cells were exposed to 10 Gy ^(60)Co γ-rays, the function of poke-weed mitogen (PWM) and ^(60)Coγ-rays irradiation effect on lymphocytes were investiga-ted by technique of ~3H-TdR incorporation. The results showed that PWM can induce both T and B lymphocytes but B lym-phocytes were more sensitive to PWM. The response of B lymphocytes to PWM wasthe same as that of non-T_4^+ cells, the response of T_4^+ cells to PWM was the lowest.DNA synthesis of both T and T_4^+ lymphocytes were synergistic to B lymphocytes.The synergistic effect in T and B lymphocytes was more strong than that in T_4^+ andB lymphocytes. When T and T_4^+ or B lymphocytes were irradiated by 10 Gy ^(60)Co γ-rays, the synergistic effect disappeared. Neither synergistic nor suppressive effect wasfound between non-T_4^+ and B lymphocytes. When T_4^+, B and non-T_4^+ lymphocyteshad been exposed to 10 Gy irradiation, the percentage suppression of ~3H-TdR incorporation between B and non-T_4^+ lymphocytes had no significant difference but was higherthan that in T_4^+ lymphocytes.
