利用SAS软件优化乙肝表面抗原压电免疫传感器的实验条件 被引量:1

摘要 目的:对聚乙烯亚胺-戊二醛交联法进行优化,提高乙肝表面抗原压电免疫传感器的灵敏度和特异性,降低生产成本,易于普遍推广。方法:利用SAS8.1软件,确定影响实验结果的主要因素、因素水平以及实验点,通过响应面分析,寻求最优值。结果:根据响应面法确定了戊二醛浓度为2.5%,PBS pH值为7.4,BSA浓度为0.5%,免疫反应时间为40 min时,信噪比达到最大,结果与预测值非常接近。结论:本实验所用四通道压电免疫传感器阵列可以同时检测两个血清样品,并且有阴阳性对照,增加了该传感器的可靠性,适合现场检测。 Objective:To optimize PEI - Glutaradehyde cross - linking method to improve sensitivity and idiosyncrasy of piezoelectric and reduce the cost, and popularize easily. Methods:Using SAS8. 1 to confirm the main factors, factor level and experimental point. Using response surface found out the best result. Results:Basing response surface confirmed that when the concentration of glutaradehyde was 2. 5%, PBS pH was 7.4, the concentration of BSA was 0. 5%, the time of immunoreaction was 40 min, the Signal -to -Noise got to biggest, and the result was closed to prediction. Conclusion:We can detect blood serumsamples at one time using four - channels piezoelectric immunosensor array. Compared with negative and positive serum, can increase the reliability of sensor. It is fit for spot detection.
