摘要
目的:探讨伊立替康联合热疗体外抑制人大肠癌细胞增殖的效果,以确定联合方案是否具有协同效应。方法:使用四甲基偶氮唑蓝(MTT)法测定单独或联合应用伊立替康与热疗对人大肠癌LOVO细胞体外增殖的抑制作用,使用流式细胞仪检测不同方案对LOVO细胞凋亡率的影响。根据Veleriote法判断热化疗联合的作用效果;透射电镜观察细胞形态。结果:与伊立替康化疗或单纯热疗作用相比,伊立替康与43℃热疗联合后对LOVO细胞增殖有显著的抑制作用(P<0.01),24h时流式细胞仪检测LOVO细胞的凋亡情况发现,热疗组、化疗组和热化疗组细胞凋亡率均较对照组显著升高(P<0.01)。结论:伊立替康与热疗联合对LOVO细胞的增殖抑制具有明显的协同作用,这可能与细胞凋亡的增多有关。
Objective To investigate the inhibitory effect of irinotecan combined with thermotherapy on the proliferation of human colon carcinoma cell line LOVO in order to learn whether the combined therapy has a synergetic effect, Methods MTY assay was applied to determine the inhibitory effect of irinotecan combined with thermotherapy, irinotecan alone, or thermotherapy alone on the proliferation of LOVO cells in vitro. Flow cytometry was used to assess the influences of the three different therapies on the apoptotic rate of LOVO cells. The effectiveness of the combined therapy was evaluated by Veleriote method. The cell morphology was observed under transmission electron microscopy. Results Irinotecan combined with thermotherapy at 43℃ significantly inhibited the cell proliferation as compared with irinotecan alone or thermotherapy alone (P 〈 0.01 ). According to the detection for 24 hours by flow cytometry, the apoptotic rates of the LOVO cells in thermotherapy group, irinotecan group, and combined therapy group were markedly higher than those in the control group (P 〈 0.01). Conclusion Irinotecan combined with thermotherapy plays a synergetic role in inhabitation of the LOVO cell proliferation, which may be associated with an increase in apoptosis of the cells.
