银杏叶提取物神经保护作用中Snk-SPAR途径机制探讨 被引量:10

摘要 为观察银杏叶提取物EGb761的神经保护作用与突触活化过程中Snk(serum-induced kinase)-SPAR(spine-associated Rap guanosine triphosphatase(GTPase)activating protein)途径之间的相关性,本研究采用RT-PCR、蛋白质免疫印迹及免疫荧光双重标记方法观察了EGb761对谷氨酸刺激后诱导的体外培养海马神经元中Snk、SPAR表达水平及分布变化的影响。结果表明,无论在mRNA水平还是蛋白质水平,EGb761均能明显阻断突触活化后Snk-SPAR途径的激活,并对谷氨酸刺激后神经元突起上Snk分布增多及其所导致的SPAR分布减少有明显的干预作用。以上结果说明,对树突棘内结构蛋白的调节以及对树突棘结构的稳定作用是EGb761神经保护作用的一个重要靶点。 In order to study the relationship between the neuroprotective effect of Ginkgo biloba extract ( EGb761 ) and Snk ( serum-induced kinase) -SPAR (spine-associated Rap guanosine triphosphatsse (GTPase) activating protein ) pathway during neuronal activity, RT-PCR, Western blot analysis, and immunofluorescence double staining were used to observe the effect of EGb761 on glutamate-induced expression and distribution changos of Snk and SPAR in cultured hippocampal neurons. The results showed that EGb761 blocked the activation of Snk - SPAR pathway and accordingly inhibited the increase of Snk and decrease of SPAR distributed in neuronal dendrites induced by glutamate stimulation. These results suggest that to regulate the expression of the structural protein in dendritic spines and stabilize the structure of dendritic spines are important targets of the neuroprotective effect of EGb761.
