摘要
前列腺素E2(PGE2)是一种致炎因子;星形胶质细胞中CREB和γ-氨基丁酸B受体(GABABR)的磷酸化,在神经系统炎症中起重要作用。为探讨PGE2对星形胶质细胞中CREB和GABABR2磷酸化的作用,本实验对体外培养的大鼠脑皮质星形胶质细胞以10μmol/LPGE2刺激后,用免疫组织化学和Western blot等方法研究了星形胶质细胞中CREB和GABABR2(Ser892)磷酸化水平在多个时相中的变化。结果显示:正常星形胶质细胞中存在着基础水平上的磷酸化的CREB和GABABR2(Ser892),经PGE2刺激培养,CREB和GABABR2(Ser892)磷酸化水平增高,并呈时间依赖趋势。结果提示,10μmol/L的PGE2能诱导大鼠脑皮质星形胶质细胞转录因子CREB发生磷酸化,也能诱导GABABR2(Ser892)发生磷酸化而影响细胞功能。由此,本文研究的结果说明PGE2可能在星形胶质细胞参与神经系统炎症的过程中发挥重要作用。
Prostaglandin E2 ( PGE2 ) has long been reported to be an important mediator in inflammation. Phasphorylation of transcription factor cAMP response clement binding protein (CREB) and GABABR2 subunit affects various ncurophysiological functions in different ways. To study the phosphorylation of CREB and GABAB R2 subunit induced by PGE2 in astrocytcs, immunocytochcmistry and Western blotting were used to determine the phosphorylation of CREB and GABAB R2 (Ser892) in cultured astrocytcs treated by 10 μmoL/L PGE2 for various periods. The results showed that basic phosphorylation levels were present for CREB and GABABR2 in normal astrocytes. The levels of phosphorylation of CREB and GABABR2 ( Ser892 ) could be observed elevated after treated with PGE2 in a time-dependent manner compared with control groups. The results demonstrate that PGE2 can induce phasphorylation of CREB and GABABR2 (Ser892) in cultured astrocytes and indicate that PGE2 may play a considerable role in inflammatory situation in central nervous system.
