结缕草属优良品系SSR指纹图谱的构建 被引量:49

摘要 采用非变性聚丙烯酰胺凝胶电泳结合快速银染检测,从45对Xgwm系列SSR引物中筛选出2对产物清晰、稳定的引物,建立了11个结缕草优良品系的DNA指纹图谱,并借鉴常规植物分类法,以品系的特征带为依据,从分子的角度将它们与生产上的主栽品种‘马尼拉’、‘青岛结缕草’和‘兰引3号’结缕草区分开来。最后利用统计软件SPSS 10.0通过欧氏平方距离法进行聚类分析,将14份参试材料分为3组。 Two SSR primers with stable amplification profiles were selected from 45 pairs of Xgwm SSR primers and the DNA fingerprint of eleven new strains of Z. japonica were established. PCR products were detected by rapid silver-stalning procedure in non-denaturing polyacrylamide gels and by DNA fingerprinting of tested materials. Conventional plant taxonomy was used for reference and these eleven strains were distinguished from the popular cultivars manila, Z. japonica cv. Qingdao and Z. japonica cv. Lanyin No. 3 at a molecular level. Finally, the accessions were classified into 3 groups by the squared euclidean distance method using SPSS 10.0 software.
