摘要
目的:探索体外诱导胎盘间充质干细胞(PMSCs)分化为神经元样细胞的方法。方法:消化胎盘组织,贴壁培养细胞,观测形态并用流式细胞仪检测其细胞表面标志;用含叔丁对甲氧酚(BHA)、二甲基亚砜(DMSO)及碱性成纤维细胞因子(b-FGF)的条件培养基培养细胞,诱导分化为神经元样细胞,采用免疫细胞化学的方法对分化的细胞进行鉴定。结果:胎盘组织的贴壁细胞与骨髓间充质干细胞(BMSCs)有相似形态和细胞表面标志,诱导后细胞表达神经元和星形胶质细胞标志神经元特异性的烯醇化酶(NSE)、和胶质纤维酸性蛋白(GFAP)。结论:胎盘组织中存在能分化为神经元样细胞的间充质干细胞。
Objective :The aim of this study is to search for the potentiality and method of inducing placenta-derived mesenchymal stem cells into neuron like cells in vitro.Methods:First,adherent culture cells were isolated from placenta entity tissue. Then these cells were examined for morphology and surface markers. Finally last the culture cells were induced to differetiate into neuron-like cells by butylated hydroxyanisole (BHA),dimethyl sulfoxide;dimethyl sulphoxide (DMSO)and basic fibroblast growth factor (bFGF), the differentiated cells were identified with immunocytochemistry. Results:Cells isolated from placenta have the similar morphology and cell surface antigen with bone mesemchymal stem cells and could be induced to differetiate into neuron-like cells, exhibit the neuonral phenotype, expressing neuron specificenolase(NSE), and glial fibrillary acid protien(GFAP). Conclusion:It is suggested that placentaderived mesenchymal stem cells can differentiate into neuron-like cells. The placenta may prove to be a useful source of MSCs.
