摘要
利用具有高活性天冬氨酸转氨酶(EC2611)的大肠杆菌(EScherichiacoli)变异株细胞,对转氨反应的过程进行了研究。测得游离大肠杆菌细胞及其固定化细胞为酶源参与的转氨反应的表观米氏常数分别为0390mol/L和0467mol/L。同时,发现辅助因子5′磷酸吡哆醛溶液预处理细胞对酶具有明显的激活作用。比较了不同的金属离子对酶的激活和抑制作用。此外,利用固定化细胞。
The results of research on the process of enzymic transamination by a mutant strain of Escherichia coli with high aspartate aminotransferase activity were reported. The apparent Michaeli′s Menten constants of free cell catalyzed and immobilized cell catalyzed transmination were determined as 0.39 mol/L and 0.467mol/L respectively. The enzymic activity can be activated by the pretreatment of pyridoxal 5′ phosphate solution and metal ion solution of Mn 2+ ,while it is obviously inhibited by metal ion Ca . The results of preparation of L phenylalanine in BSTR and PFR reactor were also reported.
