恐惧应激对雄性大鼠生殖相关参数的影响 被引量:8

摘要 目的观察比较恐惧应激状态下不同应激时间段大鼠体重、睾丸和附睾重量、精子数目、3β-羟类固醇脱氢酶(3β-HSD)蛋白表达的变化。方法建立SD大鼠的恐惧应激模型,分别取急性应激和慢性应激状态下SD大鼠睾丸、附睾称重,并对附睾尾精子计数,HE染色观察睾丸结构,免疫组织化学和western blotting分析3β-HSD表达,比较各实验组和对照组间上述指标的变化。结果急性应激组与对照组相比,大鼠体重、睾丸重量、附睾重量、精子数量没有明显变化(P>0.05),HE染色显示睾丸结构无明显变化,免疫组化显示3β-HSD表达降低,western blotting分析3β-HSD降低了7%;慢性应激组与对照组相比,大鼠体重、睾丸重量、附睾重量、精子数量都有显著差异,P<0.05。HE染色显示睾丸内生精小管细小,管腔内生精细胞稀少,免疫组化显示3β-HSD表达降低,western blotting分析3β-HSD降低了19.8%。结论急性应激没有破坏雄性大鼠机体内稳态的调节能力,对生殖的影响仅见于睾丸功能的改变,睾丸组织结构未发生变化;而长期处于应激状态下的雄性大鼠,超出机体内稳态的调节能力,对生殖的影响不仅表现在3β-HSD的表达下降,而且睾丸组织结构发生变化。提示应激是影响大鼠生殖功能的一个因素。 Objective To observe the fear-induced variation of body weight, weight of testis and epididymis, the spermatozoon number and 3 beta-hydroysteroid dehydrogenase( 3 β-HSD)expression at the different time in stressed Sprague-Dawley (SD) rat. Methods Establishment of the model of stressed SD rat was induced by fear. The testis and epididymis of SD rat in acute stress group and chronic stress group were taken respectively. The weight of testis and epididymis was measured and the number of spermatozoon from the caudal epididymis was countered. HE dyeing,immunohistochemistry and western blotting were utilized for observing the structure and the change of 3β-HSD expression in rat testis. Results Comparing with the control group,body weight, weigh of testis and epididymis,number of spermatozoon of the rats in acute stress group did not change significantly(P 〉0.05) and the structure of testicle did not changed too. The express of 3β-HSD in testis of acute stress rat reduced 7 percent. In chronic stress group, comparing with the control group body weight, weigh of testis and epididymis and number of spermatozoon of the rats were remarkable decreased (P 〈 0. 05 ) ,and the structure of somniferous tubule became thin, the cells in lumina for spermatogenesis became less and 3β-HSD expressed in rat testis reduced 19.8%. Conclusion Owing to the adjustment ability for homeostasis, the effect of acute stress on reproduction only showed functional change,such as 3β-HSD expression reduced and no significant change happed in testieular structure. On the other'hand, the chronic stress not only affected the function of testis but also affected testicular structure. These data demonstrated that stress is the one of important reason to affect reproductive function in male rat.
